7 Drug Interactions

7.1 Effects of Other Drugs on FYARRO

Additions and/or revisions underlined:

CYP3A Inhibitors and P-gp Inhibitors

Avoid concomitant use of FYARRO with strong CYP3A inhibitors and P-gp inhibitors, including grapefruit or grapefruit juice.

Reduce the dosage of FYARRO when used concomitantly with a moderate or weak CYP3A inhibitor

[see Dosage and Administration (2.3) and Clinical Pharmacology (12.3)].

Sirolimus is a CYP3A and P-gp substrate [see Clinical Pharmacology (12.3)]. Concomitant use with a CYP3A and P-gp inhibitor may increase sirolimus exposure [see Clinical Pharmacology (12.3)], which may increase the risk of FYARRO-associated adverse reactions.

Strong CYP3A Inducers and P-gp Inducers

Avoid concomitant use of FYARRO with strong CYP3A inducers and P-gp inducers.

Sirolimus is a CYP3A and P-gp substrate [see Clinical Pharmacology (12.3)]. Concomitant use with a CYP3A and/or P-gp inducer may decrease sirolimus exposure [see Clinical Pharmacology (12.3)], which may decrease the effectiveness of FYARRO.

8 Use in Specific Populations

8.6 Hepatic Impairment

Additions and/or revisions underlined:

Avoid use in patients with severe hepatic impairment.

Reduce FYARRO dosage in patients with mild or moderate hepatic impairment [see Dosage and Administration (2.4) and Clinical Pharmacology (12.3)].
